چکیده

The sex of Japanese flounder (Paralichthys olivaceus) is easily altered by water temperature or sex steroid hormone treatment during the period of sex determination. We have previously shown that rearing the genetically female larvae at high water temperature caused the suppression of P450 aromatase (P450arom) gene expression in the gonad and phenotypic sex-reversal of the individuals to males (Kitano et al. 1999. J Mol Endocrinol 23:167–176). In the present study, we show that treatment of genetically female larvae with fadrozole (aromatase inhibitor) or 17a-methyltestosterone induces sex-reversal as well as suppression of P450arom gene expression. The effect of fadrozole was counteracted by co-administration of estradiol-17b. Effective periods for fadrozole treatment to induce sex-reversal were similar to those for high water temperature treatment. RT-PCR did not detect P450arom mRNA in gonad of the sex-reversed, phenotypic males. These results indicate that sexreversal of the genetically female larvae by aromatase inhibitor (or 17a-methyltestosterone) may be due to the suppression of P450arom gene expression and the resultant decrease in the amount of estrogen. Suppression of P450 aromatase gene expression in sex-reversed males produced by rearing genetically female larvae at a high water temperature during a period of sex differentiation in the Japanese flounder (Paralichthys olivaceus).

The phenotypic sex of many teleost fishes including flounders can be experimentally altered by treating embryos or larvae with varied temperatures or sex-steroid hormones. To analyse the sex determination mechanism, especially the role of cytochrome P450 aromatase (P450arom), an enzyme that catalyses the conversion of androgens to estrogens, in temperature-dependent gonadal sex differentiation ...

Effect of Nonsteroidal Aromatase Inhibitor on Sex Reversal of Oreochromis mossambicus (Peters, 1852)

In the present study, the efficacy of letrozole, a potent nonsteroidal aromatase inhibitor, and 17α-methyltestosterone on gondal sex differentiation and sex reversal was examined in Mozambique tilapia (Oreochromis mossambicus). Among the doses tried in the experiment, 100 mg letrozole, 200 mg letrozole, and 50 mg 17α-methyltestosterone/kg produced male dominated populations (97-100% males). The...

Roles of GATA6 during Gonadal Development in Japanese Flounder: Gonadogenesis, Regulation of Gender-Related Genes, Estrogen Formation and Gonadal Function Maintenance

GATA-binding protein 6 (GATA6), a highly-conserved transcription factor of the GATA family plays an important role in gonadal cell proliferation, differentiation and endoderm development. In this study, the full-length cDNA of GATA6 of Paralichthys olivaceus (Japanese flounder) was obtained. Phylogenetic, gene structure and synteny analyses demonstrated that GATA6 of P. olivaceus is homologous ...

Roles of Two Sox9 Genes during Gonadal Development in Japanese Flounder: Sex Differentiation, Spermatogenesis and Gonadal Function Maintenance

The transcription factor sox9 has been implicated in cartilage formation and testis determination in mammals. Here, two duplicates of sox9 were found in Japanese flounder (Paralichthys olivaceus) named Posox9a and Posox9b, respectively. Phylogenetic and gene structure analyses revealed that Posox9a and Posox9b were homologous to that of teleosts and tetrapods. Quantitative real-time polymerase ...

Gonadal differentiation and effects of temperature on sex determination in southern flounder (Paralichthys lethostigma)

Southern flounder (Paralichthys lethostigma) support valuable North American fisheries and show great promise for aquaculture. Because females grow faster and reach larger adult sizes than males, monosex culture of females is desirable for commercial operations. A detailed understanding of sexual development and its timing is critical to control sex and optimize culture. Structural and cellular...
